Abstract : An adaptation and modification of the carbonatephosphotungstate technic for the determination of uric acid in rat urine is described. nEthylmaleimide is used to minimize the interfering effect of nonuric acid chromogens that contain sulfhydryl groups. Furthermore, an internal standard was incorporated to bring the uric acid values found in rat urine into an optimum range for spectrophotometric analysis. Several parameters of the basic technic were examined. Reaction mixture curves that were run on the Beckman DK-2 spectrophotometer establish maximum color development at 15 to 20 minutes, stability to 30 minutes, and an absorbence peak at 710 millimicrons. The noninterference of nethylmaleimide was demonstrated. Normal nonfasting values for rat uinary uric acid were found to be 2.17 mg./day ! 0.76 mg. (Author)
