Abstract : The dose-reduction factor (DRF) of diammonium amidophosphorothioate (A 331) has been determined in mice and related to that of cysteamine (MEA) as an internal standard. In the experiments 2.1 micro moles of MEA and 0.14 micro mole of A 331 were administered per gram of body weight. At pH 7.7 the value of DRF A 331:DRFMEA was found to be 1.15 plus or minus 0.09, and at pH 10 it was 1.08 plus or minus 0.11. The therapeutic index was 2.9 for A 331 and 1.3 for MEA. The compound A 331 was shown not to be protective at pH 3. The reasons for this finding are discussed. Serotonine injected together with A 331 does not increase the DRF over that of A 331 alone. A number of derivatives of amidophosphorothioate have been synthesized and tested, but none was found to be superior to the parent compound as a radiprotector. (Author)
