Abstract : A four year effort devoted to the establishment of fluorometric methods for the determination of acetylcholine has been completed. The use of enzymes with high specificity i.e. acetyl-cholinesterase, acetyl-CoA synthetase and citrate condensing enzyme, results in a method highly selective for ACh. At comparable concentrations, no interference is observed from acetate, acetyl-carnitine, acetyl-aspartic acid, acetyl-neuraminic acid, or from propionyl and butyryl choline esters. The method permits measurement of nanomole (10 to the -9th moles) quantities of ACh extracted from nerve tissue. In order to achieve greater sensitivity, a method describing the measurement of citrate, a final product of the acetylcholine, has been described. This method permits measurements in the picomole 10 to the -12th to -13th moles) range and is adaptable to ACH assay. Efforts will continue to combine the methods for the measurement of brain tissue ACh in the lower range. (Author)
