Abstract : In the goal of synthesizing a DNA molecule for biological studies, large amounts of protected di- and trinucleotides were required. The large scale syntheses (2-10 mmoles) of these starting blocks required chromatography through large columns of benzoylated DEAE-cellulose. This is a time-consuming and laborious step. In order to overcome this difficulty, the authors investigated the benzoylation of DEAE-Sephadex with a view to produce an adsorbent of high capacity. In this communication we wish to report the successful preparation of benzoylated DEAE-Sephadex and also its application for the large scale synthesis of protected oligonucleotides. (Author)
