Abstract : The present paper summarizes all the previous results of the authors in the area of sequential search of optimal dosages, under linear regression of dosage-toxicity. The optimal dosage is defined as the largest dosage for which the probability that the patients toxicity does not exceed a given threshold is at least gamma, 0 < gamma < 1. The search problem is a valid design problem when some (or all) of the regression parameters are unknown. In the present paper we provide a general introductory discussion concerning the applicability of the derives procedures, set up the general criteria and present the various sequential methods of on-line experimentation, which search for the location of the optimal dosage.
