Abstract : The plaque titration of yellow fever virus 17D has been reexamined according to the fundamental work of Porterfield in 1959 and 1960. The use of mixed lung and body fibroblasts of chicken embryos results in higher sensibility of the monolayers to the vaccine virus and good stainability with neutral red. Eagle basal medium combined with tris-HCl-buffer in an overlay of purified agarose afford reproducibility of plaque numbers apt to the exact checking of vaccine problems. The reading of the plaque test within five days, the comparably low costs of the cultures and the possibility of examining much larger numbers of samples offer a great advantage over the intracerebral infection of mice used at present for potency-tests of yellow fever vaccine lots. For this reason the plaque titration is recommended besides or instead of the mouse tests which last 21 days. Up to date 35 vaccine batches have been checked in parallel. (Author)
