Abstract : The report contains the results of efforts toward devising an improved method for fluorescent determination of morphine in urine. One of the initial problems investigated was a method for hydrolysis of morphine-3-glucuronide in urine without loss of morphine free base. The morphine-3-glucuronide was isolated from dog urine after intravenous administration of morphine sulfate to a beagle dog. Experiments were carried out to hydrolyze the morphine-3-glucuronide with beta-glucuronidase in solution and also with beta-glucuronidase immobilized on glass beads in a column. A number of fluorescent morphine derivatives were synthesized. These were evaluated with respect to ease of preparation, fluorescence quantum yield of product, excitation wavelength, emission, wavelength, cost of reagents and stability of reagents and product.
