Abstract : The discovery of Australia antigen (Au) and its association with serum or type B hepatitis has proven to be a major breakthrough in hepatitis research. However, the lack of a simple and economical method for isolation and purification of Au presents a major barrier to research studies in which large amounts of Au reagent are required. Electrochromatography is a new technique which combines the separation principles of electrophoresis and column chromatography. Despite its potential for separation of complex mixtures, the technique has been little used for the separation of proteins or virus particles. A simplified method for the purification of Au by electrochromatography is described which employs a new column that can be used for conventional column chromatography, preparative electrophoresis, and electrochromatography. The apparatus is unique in that it can carry out these procedures individually, in sequence, or in combination, without modification or alteration of the initial system.
