Abstract : Storage of human platelets at 4C for 24 hours results in a considerable reduction in their oxidative activity towards fatty acids, glucose and selected Krebs cycle substrates. Pursuant to these observations, the authors have focused on energy metabolism and mitochondrial behavior during platelet storage. To this end methods have been set up for measuring oxidative phosphorylation, and developing an improved method for homogenization and fractionation of human platelets. The authors have greater enrichment of mitochondria than heretofore reported. Using the density gradient method as a benchmark, a zonal ultra-centrifugation and differential centrifugation procedures will be developed for improving the purity and total harvest of mitochondria to allow studies of mitochondrial behavior during storage.
