Abstract : A simple and rapid method for the preparation of large numbers of short term cultures of erythrocytic forms of P. knowlesi is described. This method has shown itself to be valuable in testing nutritional and biochemical aspects of P. knowlesi, in testing compounds for antimalarial activity and in studying the mode of action of antimalarial drugs. 1-isoleucine and 1-methionine are essential for growth of erythrocytic forms of P. knowlesi. An extraerythrocytic source of 1-cystine, 1-tyrosine, 1-arginine, 1-glutamine, 1-histidine and 1-lysine is required for optimal in vitro development of P. knowlesi during one schizogonic cycle.
