Abstract : This report describes two methods for the determination of propyl nitrate in blood. In the first method, the propyl nitrate was isolated by addition of isopropanol to the blood and then determined in the protein-free filtrate by the 3,4-xylenol colorimetric method. Interference from chlorides in the blood was eliminated by addition of silver sulfate to the sulfuric acid reagent. Recoveries of 114 to 145 micrograms of propyl nitrate from 1 ml of blood ranged from 89% to 103% with a mean value of 97%. In the second method, propyl nitrate in the presence of 10 times as much inorganic nitrate and organic and inorganic nitrite was extracted from blood with petroleum ether and subsequently determined by the 3,4-xylenol method. The solvent extraction removed the interference from blood chloride and inorganic nitrate and nitrite. Shaking the extract with an aqueous solution of sulfamic acid or sulfanilic acid removes the interference from organic nitrite. Recoveries average 92% of propyl nitrate from a 1-ml sample of blood using sulfamic acid, and 90% using sulfanilic acid. (Author)
