Abstract : The current, standardized version of the primary mouse toxicity and gross activity screen used by the Toxicity Screening Branch of Research Laboratories, Edgewood Arsenal, is presented in this report. Essentially, the technique consists of detailed parallel observation of differences between intravenously injected control and treated mice. Under stipulated conditions, each treated animal is observed without handling and then during a battery of manipulative tests. A standard series of doses separated by specified log intervals is employed. Following injection and continuing until responses have disappeared, the routine of study is applied at 3,15,30, and 60 minutes, hourly thereafter until the end of the work day, and then daily. The method is used to determine the LD50, MED50 (the dose producing minimal effects in 50% of the animals), and a safety factor (LD50/MED50 ratio). Reactions are reported for all dose levels. This information provides a basis for recommending rejection or acceptance of a compound for further study. (Author)
