Abstract : The focal point of attack for drug design in the present investigation was to exploit the known differences of folic acid matabolism in man and the malarial parasite. It was therefore desirable to prepare target compounds in three series of nitrogen heterocycles which (a) retained antifol structural features and (b) incorporated other antimalarial moieties. The three series included are (1) tetraazanaphthalenes, (2) 1,3,5-triazanaphthalenes, and (3) 9-deazapurines.
