Abstract : During the second year of this contract, we have purified human erythrocyte acetylcholinesterase to 87% homogeneity by MAb affinity chromatography and size-exclusion HPLC and have obtained sequence data for 100 residues, derived from tryptic, chymotryptic, and V8 peptides. We have used these sequence data to synthesize oligonucleotide probes with which to screen two human cDNA libraries for the gene encoding human AChE. In addition, we have identified a large number of MAb that bind to acetylcholinesterase and we have begun to characterize several of these antibodies that are capable of inhibiting acetylcholinesterase.
