Abstract : The objective of this project was to develop radiotracer imaging technology to follow the in vivo circulation and organ deposition of liposome encapsulated hemoglobin (LEH). A liposome labeling technique based on the most common radioisotope used in nuclear medicine, technetium-99m (99m Tc), was developed and patented. A comparison of this 99mTc-liposome labeling method with previous methods using indium-111 (111 In) was performed. The label LEH was infused into small animals to monitor any in vivo differences between different LEH formulations (bovine hemoglobin, recombinant human hemoglobin, human hemolysate, lyophilized) using gamma scintigraphy and tissue biodistribution determination. These studies were correlated with any hematological and pathological changes associated with LEH treatment such as the interaction of LEH with circulating platelets using indium-111-label platelets
