Abstract : Estrogen receptor positive breast tumors are evaluated by positron emission tomography (PET) utilizing radiolabeled estrogen ligands. This imaging technique is used to predict the tumor's response to therapy and to follow the progress of disease. Desiring to find new estrogens with high binding affinity for the estrogen receptor, we synthesized 16 alpha-methoxy estradiol. This compound previously had not been isolated nor its binding affinity measured. Characterization has been achieved by 1H and 13C NMR as well as by low resolution and high resolution mass spectrometry. The synthesis was achieved by applying the chemistry of methyl hypofluorite to steroids. Optimization of reaction conditions has laid the foundation for future radiolabeling studies. Binding affinity of this compound for the estrogen receptor is currently being determined.
