Abstract : In order to better understand the extent to which the intact DNA replication machinery contributes to the overall mutation frequencies observed in normal and malignant breast cells, I have designed experiments to examine the degree of fidelity exhibited during the DNA replication process in both normal and cancerous breast cells. To accomplish this goal I have isolated a multiprotein DNA replication complex (which we have designated the DNA synthesome) from both normal breast tissue cells and malignant breast cancer cells and have begun to determine the ability of the DNA synthesome from both cell types to faithfully copy a target gene used in our in vitro replication assay system.
