Abstract : During the second year of this grant, we obtained better X-ray data from the crystals of the T-cell receptor/HLA-A2/peptide complex, by finding improved conditions for freezing the crystals, by producing heavy atom-containing crystals, and by collecting data at the Cornell High Energy Synchrotron Source (CHESS). By the combination of the techniques of molecular replacement, multiple isomorphous replacement, and iterative real-space averaging, we were able to determine the structure of the TCR/HLA-A2/peptide complex. We are in the process of analyzing the immunological implications of the structure and are determining two closely related structures to allow a fuller interpretation of this central molecular recognition event in the immune system.
