Abstract : Mouse mammary tumor virus is used as a mutagen in transgenic mice that carry an activated Wnt-1 gene, to produce additional events such as activation of a second oncogene by nearby insertion of proviral DNA. Cloning cellular sequences flanking a new proviral insertion from tumor 76, identified a common insertion locus for MMTV. Further analyses of this region identified the proto-oncogene Fgf3 to be activated over a long range by the MMTV insertions. Fgf8 is another proto-oncogene that we identified to be activated by MMTV in 10% of these tumors. Characterization of the oncogenic potentials of Fgf8 isoforms in mammary epithelial cells showed a surprising finding; the mammary epithelial cells stimulated by Fgf8b protein containing conditioned medium went through programmed cell death or apoptosis.
