Abstract : Serum-free media were developed for the growth of diploid fetal Rhesus monkey lung (DBS-FRhL-2) cells and for the propagation of dengue type 2 virus, vaccine strain (dengue-2 vaccine virus). The serum originally required in the medium was substituted with a combination of vitamins, amino acids, growth factors, hormones, other organic compounds and inorganic salts. Three other cells were adapted to grow serially in serum-free medium: 1) rhesus monkey kidney cells (MK-2, LLC-1) using our WO5 medium in shaker and monolayer cultures; 2) human lung and 3) human foreskin cells (diploid) in WO20 medium in monolayers. There was not sufficient time and funds to test these cells for their susceptibility and determination of peak titer of dengue-2 parent and vaccine viruses.
