Abstract : This report describes synthetic work performed in a search for new prophylactic antiradiation drugs with emphasis on the quest for an effective agent capable of being administered by mouth. Accordingly, compounds selected for the synthetic efforts were designed to incorporate the structural features of effective agents that require injection (such as WR-2721) with other structural features that might promote effectiveness following oral ingestion. Toward this aim we designed several analogs of WR-2721 bearing carboxyl substituents on the rationale that absorption of such compounds might occur by the same mechanism as that attending amino acids in nutritional processes. Multistep syntheses of three carboxy-substituted analogs of WR-2721 were satisfactorily completed as were key steps toward another. Two compounds of the WR-2721 class bearing adamantyl groupings on terminal nitrogen were also prepared. Synthesis of a new phosphorothioate, S-2-(4-PIPERIDYLMETHYL aminoethyl dihydrogen phosphorothioate, proceeded well through the key intermediate. Plans to prepare thiazolidine derivatives of known aminothiol radioprotectors were initiated in preliminary experiments involving L - 2, omega-diaminoalkanethiols.
