Abstract : A set of assays have been developed in which cell cultures serve as an alternative to in vivo toxicity testing. One test uses a morphological screening procedure based on the highest dose of toxic agent tolerated (HTD) and is followed by a quantitative neutral red spectrophotometric absorption assay (NR-90). The method has been standardized to make use of an automated microtiter plate reader. A second test is based on 50 percent inhibition of tritium uridine uptake (UI-50) after incubation of the cultures with the toxicant. There is good agreement between these assays in the rank correlation of a broad spectrum of agents tested, as well as with the in vivo Draize rabbit eye irritancy data.
