Abstract : Human geneticists have been extremely successful in the past decade in mapping rare disease genes. For common diseases with a substantial genetic component, the payoff for human health is larger, but the mapping problems are harder. There is a need for robust statistical techniques that require minimal assumptions about the mode of inheritance of the disease studied. The affected-pedigree-member (APM) of linkage analysis makes virtually no assumptions about disease transmission except that it is independent of marker transmission in a pedigree. We discuss here an extension of the APM method from single markers to multiple closely linked markers. This extension should improve the power of the APM method to detect linkage.
