Abstract : Both urokinase (UK) and t-PA are major fibrinolytic enzymes widely used as efficient anti-thrombosis agents. The t-PA induced plasminogen activation can be strongly stimulated by fibrin. It is due to the binding both of t-PA and Glu-plasminogen to the D-domain of fibrin. The fibrin A alpha 149-161 sequence had been identified as a determinant for this stimulation. By contrast, UK (two-chain form) displayed little fibrin stimulation. A similarity of charge and hydrophobic distribution between UK 149-158 and A alpha 149-161 was found with a pattern-recognition program. The present study was undertaken to understand the structure-function relationship of UK 149-158.
