Abstract : The hypervariability of the gp120 envelope protein principal neutralizing domain, the V3 loop, represents a major problem in the design of vaccines against HIV-1. In this region, differences as high as 50% can be found between isolates. Recombinant proteins and peptides derived from different isolates have been shown to induce mostly HIV-1 subtype-specific neutralizing antibodies. Broadly reactive, neutralizing, antibodies have been obtained using discrete mixtures of different peptides or hybrid V3-peptides. However, even though some cross-reactivities may be expected, point mutations giving rise to new variants escaping neutralization are still likely to occur. An ideal solution for a vaccine would be to recruit simultaneously the broadest part of the immune repertoire capable of recognizing not only the known isolates but also the highest possible number of escaping mutants.
